چکیده

The coactivator PGC-1α is a key regulator of mitochondrial biogenesis and respiration, mediating expression of several transcription factors required for these programs. Three new studies reveal that PGC-1α expression is downregulated in patients with Huntington’s disease (HD) and in several animal models of this neurodegenerative disorder, implicating PGC-1α in HD pathogenesis and providing a connection between impaired energy metabolism and neurodegeneration (Cui et al., 2006; Weydt et al., 2006; St-Pierre et al., 2006).

BIOLOGICAL SCIENCES: Neuroscience PPARγ-coactivator-1α gene transfer reduces neuronal loss and amyloid-β generation by reducing BACE1 in an Alzheimer’s disease model Short title: PGC-1α gene therapy for Alzheimer’s disease

Current therapies for Alzheimer’s disease (AD) are symptomatic and do not target the underlying Aβ pathology and other important hallmarks including neuronal loss. PPARγ-coactivator-1α (PGC-1α) is a transcriptional regulator of metabolic genes, oxidative phosphorylation and mitochondrial biogenesis. We previously reported that PGC-1α also regulates the transcription of BACE1, the main enzyme in...

Skeletal muscle SIRT1 and the genetics of metabolic health: therapeutic activation by pharmaceuticals and exercise

Silent mating type information regulation 2 homolog 1 (SIRT1) is implicated in the control of skeletal muscle mitochondrial content and function through deacetylation of peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α) and participation in the SIRT1/PGC-1α axis. The SIRT1/PGC-1α axis control of skeletal muscle mitochondrial biogenesis is an important therapeutic target for o...

Mitogen- and stress-activated protein kinase 1-induced neuroprotection in Huntington's disease: role on chromatin remodeling at the PGC-1-alpha promoter

Huntington's disease (HD) is a neurodegenerative disorder due to abnormal polyglutamine expansion in huntingtin protein (Exp-Htt). This expansion causes protein aggregation, leading to neuronal dysfunction and death. We have previously shown that mitogen- and stress-activated kinase (MSK-1), a nuclear protein kinase involved in chromatin remodeling through histone H3 phosphorylation, is deficie...

PGC-1α activity in nigral dopamine neurons determines vulnerability to α-synuclein

INTRODUCTION Mitochondrial dysfunction and oxidative stress are critical factors in the pathogenesis of age-dependent neurodegenerative diseases. PGC-1α, a master regulator of mitochondrial biogenesis and cellular antioxidant defense, has emerged as a possible therapeutic target for Parkinson's disease, with important roles in the function and survival of dopaminergic neurons in the substantia ...

Transducer of regulated CREB-binding proteins (TORCs) transcription and function is impaired in Huntington's disease.

Huntington's disease (HD) is an incurable neurological disorder caused by an abnormal glutamine repeat expansion in the huntingtin (Htt) protein. In the present studies, we investigated the role of Transducers of Regulated cAMP response element-binding (CREB) protein activity (TORCs) in HD, since TORCs play an important role in the expression of the transcriptional co-regulator peroxisome proli...
